Similarly to yesterday, Rea had a better getaway and was ahead of Sykes at Turn 1, the latter dropping behind Alex Lowes as well.
However, he fought back and retook the lead a corner later, sneaking past on the outside of both Rea and Lowes.
In the lower parts of the top 10, Markus Reiterberger touched the back of Chaz Davies and flew several metres in a highside crash, from which he walked away limping.
A lap later Chaz Davies lost control of his bike and, running right behind the Welshman, Michael van der Mark failed to avoid him and crashed too.
The Kawasaki duo soon pulled away from the rest of the field, with Lowes narrowly avoiding a crash after a wobble and falling behind the Ducatis of Davide Giugliano and Xavi Fores.
Having closely followed Sykes throughout the race, Rea made his move five laps before the end, forcing his way through on the outside of Turn 1.
Unable to fight back, Sykes quickly fell more than a second behind Rea, the latter celebrating his third double win of the season.
Giugliano took a podium on home soil, followed by Fores and Lorenzo Savadori, who took advantage of a late-race problem for Lowes.
The Briton ended up eighth, also allowing Nicky Hayden and Jordi Torres through.
His Yamaha teammate Niccolo Canepa finished right behind in ninth with Van der Mark recovering from last to 10th.
