It took Vinales five minutes to beat Danilo Petrucci's FP1 benchmark of 1m34.680s, but Folger soon took over with a 1m33.898s.
The first runs were interrupted by rain around 15 minutes into the session, but the field was able to re-enter a dry track just six minutes later.
Vinales soon reclaimed first place by going 0.077s quicker but, with rain predicted for Saturday, the session ended with a shootout as everyone tried to secure a place in the top 10.
Pramac Ducati's Petrucci and Tech 3 rider Johann Zarco both came close to demoting Vinales, but it was Marc Marquez who managed to lower the benchmark.
However, Vinales immediately fought back with a 1m33.696s and, unlike Marquez, improved with his next flyer as well by half a second.
His 1m33.130s time ended up the fastest lap of the session, with his closest challenger Folger being 0.367s off the pace.
Marquez's Repsol Honda was a further three tenths down in third, ahead of Ducati rider Andrea Dovizioso, who used the bike that broke down at the beginning of FP1.
Cal Crutchlow put a second Honda in the top five, ahead of Valentino Rossi, who jumped up to sixth with a late flyer.
Petrucci, Zarco, Aspar Ducati's Alvaro Bautista and Dani Pedrosa took the final positions in the top 10.
Bautista's teammate Karel Abraham missed out on a potential automatic Q2 spot by 0.151s in 11th, while Jorge Lorenzo had a low-key session in 14th behind Aprilia's Aleix Espargaro.
Espargaro's teammate Sam Lowes had a crash at Turn 10 towards the end of the session, the Briton ending up 20th.
