Following the cancellation of Sunday's morning's planned extra practice due to poor weather, warm-up was extended from its usual 20-minute run time to 30-minutes, and took place on a wet Termas de Rio Hondo track.
Iannone was the man to beat throughout much of the session, becoming the first rider to dip beneath the 1m50s with a benchmark of 1m49.518s 10 minutes in that stood until the closing stages.
Works Honda rider Dani Pedrosa beat the Italian by three tenths with a minute to go until the chequered flag with a 1m49.231s, which was fractionally bettered by teammate Marc Marquez after the clock hit zero.
However, Iannone's final effort of 1m49.177s was good enough to depose his former Moto2 rival by 0.030s, with Pedrosa slotting back into second with a time just 0.006s slower.
Hector Barbera was an impressive fourth on the Avintia Ducati, just 0.082s off the pace, with Valentino Rossi completing the top five on the best of the Yamahas, 0.279s off the pace.
Andrea Dovizioso (Ducati), Yonny Hernandez (Aspar Ducati), Jack Miller (Marc VDS Honda), Cal Crutchlow (LCR Honda) and Barbera's Avintia teammate Loris Baz completed the top 10, with reigning champion Jorge Lorenzo lapping 1.876s off the pace in 12th.
All eyes will now be on the weather conditions, as a dry track for the race would now mean a reduced race distance of 20 laps and a mandatory bike swap.
