After Andrea Dovizioso posted the quickest time on Tuesday - a day likewise dominated by the Italian marque - Iannone lowered the benchmark slightly in Wednesday's morning session, before improving massively in the closing minutes of the afternoon.
Iannone's final time ended up a 1m23.240s, while none of the non-Ducati bikes even managed a sub-1m24s lap.
In the morning session, Iannone dethroned Dovizioso with less than half an hour to go and would shave off a couple of hundredths of a second a few laps later, ending up at 1m23.719s.
Iannone would then make it a 1m23.662s in the afternoon session, before lowering the benchmark by four tenths with less than five minutes remaining.
Dovizioso wound up second while Casey Stoner, looking set to improve in the final moments of the session, ended up crashing out and settling for third.
As such, with three factory Ducatis and the satellite Avintia of Hector Barbera locking out the top four, Valentino Rossi had to do with being best of the rest - his late 1m24.169s effort deposing Maverick Vinales as the quickest non-Ducati rider of the test.
Jorge Lorenzo moved up shortly thereafter, ending the test 0.025s off his teammate.
Vinales and Suzuki teammate Aleix Espargaro were seventh and eighth overall on the day, with the top-10 made up by two more Ducatis - Pramac's Scott Redding and Yonny Hernandez, whose best time had put him third in the morning session.
The series' new manufacturer KTM ended the test a little under two seconds off the pace.
